The dataset share information for 13 specimens examined during the description of the new Suillus species from North Asia.The specimens of the Suillus bresadolae f. flavogriseus (Cazzoli & Consiglio) Klofac, Suillus nueschii var. caerulescens Singer, Suillus gastroflavus Zvyagina, Rebriev, Sazanova & E.F. Malysheva and new species are annotated. Annotations are accompanied by geographic data, images of original labels, fruiting bodies, exsicсates and microstructures.

The new laboratory of molecular methods for biodiversity research and monitoring was organized thanks to the support of the Russian Ministry of Sience and Education grant for organization of New Young Researcher Laboratories as part of the implementation of the national project Science and Universities.

Descrição do Design Our goal is to develop molecular methods of biodiversity inventory with the aim of environmental impact assessment in the region of intence oil and gas management. With special love for fungi and raised bogs, we intend to initiate the molecular approach to study these important and vulnеrable communities

Métodos de Amostragem

Collections were made, documented and preserved using the standard methods (Bondartsev, Zinger, 1955, Tulloss, 1999, Halling, 1996)

Área de Estudo The dataset contains annotations of specimens used for comparative and phylogenetic study, as well as their images and links to DNA sequences in the GenBank
Controle de Qualidade All determinations could be checked online or by the request of specimens from the LE and MAG. DNA sequences are deposited in Genbank.

Descrição dos passos do método:

  1. Specimens were photographed in situ, examined for taste and smell, dried at temperatures below 50°C.
  2. Macroscopic descriptions were based on the study of both fresh and dried materials as well as on photographs.
  3. Microstructures were observed at ×400 and ×1000 in squash preparations in 5% KOH, Congo Red, and Melzer’s reagent. Up to 30 basidiospores, 10 hymenial cystidia, and 10 terminal elements of the pileipellis per specimen were measured to obtain descriptive statistics.
  4. Measurements were made in ADFImageCapture calibrated by OMP object-micrometre (LOMO). Dimensions are given as (absolute min) average min–average max (absolute max) of the all examined specimens, Q = average min–average max quotient (length/width ratio).
  5. PCR ITS products were obtained without DNA extraction using the standard protocol of Thermo Scientific Phire Tissue Direct PCR Master Mix kit.
  6. For amplification of the ITS region the primers ITS1-F, ITS4-B (Gardes, Bruns, 1993) were used.
  7. LSU (28S) was amplified with LR0R, LR5 (Vilgalys, Hester, 1990) pair of primers, TEF1 with EF1-983F and EF1-1567R (Rehner, Buckley, 2005).
  8. Amplified products were sequenced using BigDyeH Terminator 3.1 Cycle Sequencing Kit (Applied Biosystems, Foster City, California).
  9. The sequences were assembled in CodonCode Aligner V.9.0.1 (CodonCode Corporation).
