Descrição
Elena Zvyagina's personal collection of fungi collected as a result of field research in various regions of Russia. At the moment, the dataset is loaded mainly with specimens collected in North Karelia, Western Siberia, and the Russian Far East. The collection dataset also includes parts of specimens loaned by various herbaria and personal collections in the course of work on Boletaceae, even if these specimens were returned to the herbarium. The classical method of collection and preservation of dry specimens of macrofungi is used to store the material. A large part of collection has been sequenced and has attached sequence material. The collection is stored at the White Sea Biological Station of Moscow State University (186672, Russia, Republic of Karelia, poselok Primorsky).
This IPT archives the data and thus serves as the data repository. The data and resource metadata are available for download in the downloads section. The versions table lists other versions of the resource that have been made publicly available and allows tracking changes made to the resource over time.

Dados Sobre o Projeto
The new laboratory of molecular methods for biodiversity research and monitoring was organized thanks to the support of the Russian Ministry of Sience and Education grant for organization of New Young Researcher Laboratories as part of the implementation of the national project Science and Universities.
Título | Laboratory for the development of a metagenomic analysis methodology for express assessment of environmental impacts in conditions of intensive subsoil use on the basis of Yugra State University |

Financiamento | Russian Ministry of Sience and Education grant for organization of New Young Researcher Laboratories as part of the implementation of the national project Science and Universities |
Descrição da Área de Estudo | Our goal is to develop molecular methods of biodiversity inventory with the aim of environmental impact assessment in the region of intence oil and gas management. With special love for fungi and raised bogs, we intend to initiate the molecular approach to study these important and vulnеrable communities |
O pessoal envolvido no projeto:
Métodos de Amostragem
Specimens were photographed in situ for macromorphological features. Fading out features like smell and taste were described in field. Sporocarps were dried in the mushroom dryer under 40-50 °C. Dried specimens are stored in Ziplocs and paper envelopes.
Área de Estudo | Collections were made mostly in boreal zone of Eurasia during the study of fungal diversity of taiga forests, subpolar mountains ecosystems and taxonomy of Boletales order. |
---|---|
Controle de Qualidade | Most of the specimens are providet with photografs. DNA sequences uploaded to GenBank. Specimens are available by the personal request |
Descrição dos passos do método:
- Forays were undertaken randomly
- Specimens were photographed in situ for macromorphological features
- Fading out features like smell and taste were described in field
- Sporocarps were extracted from substrate and putted into aluminum foil
- Sporocarps were dried in the mushroom dryer under 40-50 °C
- Dried specimens are stored in Ziplocs and paper envelopes
- For amplification of the ITS region the primers ITS1-F, ITS4-B (Gardes, Bruns, 1993) were used
- LSU (28S) was amplified with LR0R, LR5 (Vilgalys, Hester, 1990) pair of primers, TEF1 with EF1-983F and EF1-1567R (Rehner, Buckley, 2005)
- Amplified products were sequenced using BigDyeH Terminator 3.1 Cycle Sequencing Kit (Applied Biosystems, Foster City, California). 10) The sequences were assembled in CodonCode Aligner V.9.0.1 (CodonCode Corporation)
